The prevalence of obesity has surged worldwide since 1975, constituting a major global health crisis. While lifestyle factors such as diet and exercise contribute to obesity, researchers from the University of California San Diego School of Medicine have uncovered new insights into how obesity impacts our mitochondria, the cellular structures responsible for energy production.

Published on January 29, 2023, in Nature Metabolism, the study reveals that mice fed a high-fat diet experienced fragmentation of mitochondria within their fat cells, resulting in smaller mitochondria with reduced fat-burning capacity. Remarkably, the researchers identified a single gene controlling this process. Deleting this gene in mice protected them from excessive weight gain, even when consuming the same high-fat diet as their counterparts.

Everyone knows that overeating can lead to weight gain. However, it also triggers a metabolic “domino effect”, reducing your ability to burn calories, making the situation worse, and spiraling out of control.

Obesity, affecting over 40% of adults in the United States, arises when the body accumulates excess fat, primarily stored in adipose tissue. This tissue serves vital functions such as cushioning organs, providing insulation, and regulating metabolic processes. However, in cases of caloric imbalance like obesity, the ability of fat cells to burn energy diminishes, posing challenges for weight loss.

To investigate this phenomenon, the researchers fed mice a high-fat diet and monitored its effects on mitochondria within their fat cells. They observed mitochondrial fragmentation, wherein the structures split into smaller, less effective mitochondria following the high-fat diet. Furthermore, they identified RaIA, a molecule with multiple functions, including the breakdown of malfunctioning mitochondria, as the driver behind this metabolic alteration associated with obesity.

What’s more, chronic activation of this RaIA seems to play a key role in destroying energy expenditure in obese adipose tissue – the hardest fat to get rid of. A proper understanding of this fat-burning mechanism makes it easier for medical professionals to come up with more targeted therapies to confront weight gain and related metabolic issues by increasing the body’s ability to burn fat.

Deleting the gene linked to RaIA protected mice from diet-induced weight gain, suggesting its potential as a therapeutic target. Additionally, the researchers found similarities between the proteins affected by RaIA in mice and those associated with obesity and insulin resistance in humans, implying shared mechanisms driving human obesity.
